Jujube seed, also known as red date, is used in Eastern traditional medicine for various traits. The Japanese Pharmacopoeia defines jujube seed as the seed of Ziziphus jujuba. However, most of the jujube seed in Japan is imported from China and other Asian countries, and recent DNA studies have shown that imported jujube seed originates from Z. mauritiana as well as Z. jujuba. A method for distinguishing "true" jujube seeds from Z. mauritiana seeds using a chemical approach is thus desirable.
For this reason, a Japanese research team sought to establish a purity test for jujube seed. They identified Jujuboside A as a compound specific to Z. jujuba, and frangufoline and oleanolic acid as marker compounds specific to Z. mauritiana. Thin-layer chromatography (TLC) and gas chromatography-mass spectrometry analyses revealed that the latter compound was useful for testing by TLC analysis. Fractionation was performed on an Isolera™ Dalton with a 25 g Biotage® SNAP Ultra cartridge.
